Before painting, make sure the wood has been thoroughly primed with gesso or an appropriate primer. 

Yes, you can paint over an existing artwork on any surface but with few precautions; sand the the surface before painting over to improve the adhesionApply a primer or gesso for smoothness, to make your surface a solid foundation for your new artwork.
